Not specifically Amazon, but it depends on what type of design: UI/UX, web, marketing, print/packaging? When I interview folks, beyond awesome technical skills and portfolio, I look for having a well thought out process that incorporates feedback touchpoints from all stakeholders and ability to work as a team. From lo-fi to hi-fi, all the little things should be systematically worked out in their process. They should have the ability communicate and present concepts in a way that doesn’t require intensive verbal explanations. Let the work speak for itself. Is this perhaps an initial phone screen or actual interview? If the former, they may ask about your career goals and what you’ve done so far (Be confident, but not cocky. We can always learn new things.) It’ll probably lead to a small design/technical test, if they like what they hear, that may result in a formal video interview and call back.
