Officially, Asus lists 3 ways to update BIOS: two methods need a FAT formatted USB drive, the third method needs AI Suite 3 / EZ Update software. I've found a fourth method than needs neither a USB drive nor any Asus software/bloatware installed, and is actually the easiest to use.
My method uses EZ Flash 3 utility that is baked into UEFI BIOS, but it doesn't need a USB drive to read the new bios update file. What Asus doesn't say and what I accidentally found out it that EZ Flash 3 can read NTFS drives. You can simply download the bios file to your main C:/ (Windows) drive and update from there. Here's are the instructions in detail:

Download the latest BIOS update from ASUS support page for your specific motherboard.

After unzip, a .CAP file will appear, this is the BIOS update file. OPTIONAL: Move this file somewhere where it's easy to navigate, such as C:\.

Restart computer and press F2 to enter BIOS. Select Advanced Mode (F7), go to Tool > ASUS EZ Flash 3 utility. Under Storage Device(s) locate your main drive (It's usually the biggest one). Navigate to the .CAP file and click YES/OK to confirm update.
The bios flashing last about 5 minutes. When the progress bar is completed, click OK to reboot. That's it.
Needless to say, this method will not work if your drive is encrypted or if you have older non-UEFI bios motherboard that doesn't have Flash 3 utility.
It works for me on my B550 mobo and Windows. Let me know if it works for you.
I have an ASUS Maximus ROG XIII Hero with BIOS v0707 and would like to update to the latest version (1301) but I’ve never updated a BIOS before.
