So I've set up an artificial jungle biome to farm jungle spores and other stuff, but no spores seem to be spawning.

I know the biome is valid because the music and background change. Jungle plants spawn and even some hornets and slimes will spawn....i even got a Plantera bulb growing there and i've found a couple of life fruit, but still not a single spore.

Anyone have any advice? Do spores have to be grown in deeper jungle?

It depends on where you go in the jungle. The areas closer to top level do not have too many. As you explore more and more and reveal more areas, you will find more. You also should learn how to mine in water, in case you do not have something that will help you breathe under water. Even on subsequent visits to the same area, I see more spores have grown, and I get those also.
