Super easy, make sure the app is set up to have the home screen shortcut where a little bar appears on the bottom of the screen, then make sure you have a card already set up in the app before you go to a store. Once that's all good and ready to go all you need to do is go to your home screen when you're checking out at the register, swipe up on the little bar, and it'll either ask for your PIN or if you've set up the fingerprint you can just use that, and it'll vibrate and show a countdown. Once that's happened you just tap and hold the phone against the credit card reader. If you're using a case that's too thick for wireless charging it'll probably be too thick for the MST mode on Samsung Pay but it should still work with any Tap-to-pay readers using NFC.

personally I really don't like the gesture where you swipe up for the card. you set up your credit card in the app (you'll likely have to speak on the phone with your bank which only takes a few minutes). then once you're setup, anytime you're at an NFC or even a regular credit card terminal you just pull out the app select the card and use either your irises or your fingerprint to authenticate for the purchase. it's extremely quick and easy and the receipt shows up immediately on your phone. I can't recommend it enough, I don't even carry my credit card around anymore.
