I live in Canada, if I buy a Spotify Premium Gift Card from Target, and redeem it on my Spotify Account that I use a VPN on, will it work?

You can redeem e-cards in any account. I'm sure physical cards will do the same.

I live in Turkey, and Spotify isn't available in my country. Except Premium. But I can't buy Premium without a bank account in "available" countries- not even with PayPal.

So what I do is to ask one of my friends who lives in USA, to buy me an e-card gift. Then I can activate it without even using VPN or anything to fake my location.
