Yesterday I introduced my mother to Hamilton and she watched it for the first time. We're from Germany so she didn't understand everything perfectly. When she asked me what the line "I am not throwing away my shot!" means, I didn't really know how to explain it to her, so if anyone of you has a good explanation please tell me! Thanks in advance :D

The line kind of hinges on a double meaning. The first meaning is the colloquial English term "throw away one's shot," referring to wasting an opportunity. Hamilton is saying that he will not waste the opportunity he has been given, but will instead pursue his dreams. The second meaning, more particular to the play, refers to the practice of "deloping," in which a duelist intentionally wastes their shot in order to bring an end to the duel without either party being injured. In this sense, "throwing away my shot" is literal; Hamilton "throws away" (wastes) the shot from his pistol.
